PENGARUH ZAT PENGATUR TUMBUH BENZYL ADENIN ( BA ) DAN NAA TERHADAP PERTUMBUHAN TEMU LAWAK ( CURCUMA XANTHORHIZA ROXB. ) SECARA IN VITRO

ABSTRAK

Penelitian perbanyakan temu lawak secara in vitro telah dilakukan di Laboratorium Kultur Jaringan Kelti Plasma Nutfah dan Pemuliaan,Balai Penelitian Tanaman Rempah dan Obat, Bogor mulai bulan Oktober 1999 sampai Juli 2000 yang bertujuan untuk mengetahui pengaruh beberapa taraf konsentrasi benzyl adenin (BA) dan NAA terhadap pertumbuhan temu lawak secara in vitro. Bahan tanaman yang digunakan adalah mata tunas temu lawak dari pertanaman di Instalasi Penelitian Sukamulya, Sukabumi. Sebelum di kultur pada media perlakuan,mata tunas disterilkan dengan menggunakan sterilan alkohol 70 %, HgCl 2 0.2 %, Clorox 20 % dan terakhir dibilas akuades steril 3 kali. Murashige dan Skoog (MS) yang diperkaaya dengan vitamin dari group B digunakan sebagai media dasar. Beberapa taraf konsentrasi zat pengatur tumbuh benzyl adenin (BA) yaitu 0; 0.5; 1.0; 1.5 dan 2.0 mg/l secara tunggal serta kombinasi dengan NAA 0.5 mg/l digunakan sebagai perlakuan. Rancangan yang digunakan adalah acak lengkap dengan tiga ulangan dan setiap ulangan terdiri atas lima botol. Parameter yang diamati adalah rata-rata jumlah tunas, panjang tunas, jumlah daun dan akar serta penampakan biakan secara visual. Hasil penelitian menunjukkan bahwa dalam waktu 8 minggu, jumlah tunas dan daun terbanyak diperoleh pada perlakuan BA 1.5 mg/l . Akar terbanyak diperoleh pada perlakuan BA 1.0 mg/l + NAA 0.5 mg/l. Penggunaan BA secara tunggal maupun kombinasi dengan NAA tidak berpengaruh nyata terhadap tinggi tunas. Kedua zat pengatur tumbuh BA maupun kombinasi dengan NAA dapat digunakan untuk perbanyaka n temu lawak secara in vitro.

Effect of Benzyl Adenin (BA) and NAA As Growth Regulatur On The Growth Of Temoe Lawak (Curcuma xanthorriza Roxb.) Through In Vitro

ABSTRACT

Micropropagation of temoe lawak in vitro has been conducted in Tissue Culture Laboratory Germplasm and Breeding Division at Research Institute for Spice and Medicinal Crops,Bogor from October 1999 to July 2000. Shoots of temoe lawak from Sukamulya Research Instalation were used as explants. Shoots were sterilized with sterilant such as 70 % alcohol , 0.2 % mercuri clorite , 20 % Clorox and then cleaned with aqudes before they are cultured. Murashige and Skoog (MS) which was added with vitamine from B group were used as basic medium. Several concentration of benzyl adenin i.e : 0; 0.5; 1.0; 1.5 and 2.0 mg/l singly and combination with NAA 0.5 mg/l were used as treatments. The treatment were arranged in Randomized Completely Design with three replications. Each replication consited of five bottles. The parameters observed were average the number and lengths of shoot , number of leaves and roots and the culture performance visually. The result showed that the greatest number of shoot and leaves were found by using 1.5 mg/ l BA during 8 weeks. The greatest roots were found on 1.0 mg/l BA + 0.5 mg/l NAA . BA used singly or combined with NAA could be used on micropropagation of temoe lawak in vitro.

EVALUASI BEBERAPA SIFAT AGRONOMI PLASMA NUTFAH TEMU LAWAK (Curcuma Xanthorriza Roxb.)

ABSTRAK

Sejumlah 19 nomor plasma nutfah temulawak telah ditanam di Instalasi Penelitian Sukamulya (Sukabumi), pada tahun 1998/1999. Setiap genotipa ditanam sebanyak 5 baris dengan jarak tanam 60 x 60 cm. Masing-masing baris terdiri dari 10 rimpang induk. Pemupukan menggunakan urea, SP-36 dan KCl diberikan pada saat tanam. Hasil penelitian menunjukkan adanya variasi pada beberapa karakter agronomi yang diamati yaitu tinggi tanaman berkisar antara 97 – 184 cm; jumlah anakan berkisar antara 3 – 7.1; jumlah daun berkisar antara 6.6 – 8.7; panjang daun 55.1 – 101 cm; lebar daun 16.5 – 25.2 cm; diameter batang antara 7.8 – 13.9 mm; rata-rata berat rimpang per rumpun berkisar antara 1.2 – 4.2 kg. Rata-rata produksi rimpang per rumpun di tingkat petani berkisar 2 – 2.5 kg. Diperoleh 10 nomor plasma nutfah yang memiliki potensi produksi tinggi dengan produksi rimpang lebih dari 2.5 kg per rumpun.

Evaluasi On Some Agronomic Characters Of Temulawak Germplasm (Curcuma xanthorriza Roxb.)

ABSTRACT

A total of nine teen temulawak germplasm were evaluated at the Research Instalation Sukamulya (Sukabumi) during the wet season of 1998/1999. Each genotype was palnted in five rows, each consisted of 10 rhizomes. Plant spacing was 60 x 60 cm. The plants were pertilized at the rate of urea, SP-36 and KCl per rhizome given at planting time. Result showed that there were variation on some agronomic characters, i.e. plant height range of 97 – 187 cm; number of tiller range of 3 – 7.1; number of leaves range of 6.6 – 8.7; length of leaves range of 55.1 – 101 cm; width of leaves range of 16.5 – 25.2 cm; the diameter of stalk range of 7.8 – 13.9 mm; and average yield of rhizome per individual plant range of 1.2 – 4.2 kg. The average production of rhizome at farmers level as 2 – 2.5 kg/plant. The result of the evaluation showed that there were ten numbers of germplsm possessive yield potensial of more than 2.5 kg per individual plant.
